Job Isn't Posted Yet, How Do I Prepare to Apply?

Dear Experts, A job is opening up, but I'm not supposed to know. What can I do to prepare to apply? I know someone is leaving in our department. I think her position will be filled. I want to apply before they post it. I'm not supposed to know she is leaving though. What can I do in advance to prepare?
